Abstract

In this chapter, a novel method has been developed for simultaneous estimation of Clobetasol, Neomycin, Chlorhexidine and Miconazole in bulk and marketed formulations. Chromatography was carried out using a Water's 2695 HPLC system equipped with a Hamilt syringe, an auto sampler, and a 2996 photodiode array detector. The best outcomes were achieved using phosphate buffer (20 Mm), pH 6.6 adjusted with diluted orthophosphoric acid, Acetonitrile taken in Gradient Program at flow rate of 1 ml/m, and detection at 239 nm. For all four drugs, the retention time was less than 9 minutes. The method was demonstrated to be linear, precise, and accurate in accordance with ICH guidelines.

The findings showed that the linearity ranges between 1.25 and 300 µg/ml. The LOQ values were discovered to be 0.05, 0.23, 0.11, and 1.20. The sample solution injected after 24 hr did not show any appreciable change.

The developed method was simple, specific, and sensitive because the excipients did not interfere with the determination of the main components. The proposed technique can be used for routine analysis of Clobetasol, Neomycin, Chlorhexidine, and Miconazole in combined dosage form, that are available in variable concentrations.
